ABSTRACT

Isocorydine and its analogs were extracted from Dicranostigma leptopodum and Stephania yunnanensis through the method of natural products chemistry. Its derivatives were prepared by chemical structure modifications from isocorydine. MTT method was used to study the inhibitory effect of those compounds on the growth of HepG2, HeLa and MGC-803 cancer cell lines in vitro. The results showed that isocorydine and its analogs all have the growth inhibition for those cancer cell lines. This paper investigated the structure-activity relationship of isocorydine and its derivatives with anticancer activity in the aspect of stereochemical structure, functional groups positions of the compounds and the electron density of aromatic rings based on the single crystal diffraction structure and the molecular docking of EGFR and isocorydine.

ABSTRACT

Objective To investigate the alterations of tight junction proteins in duodenal mucosa from 6-hydroxydopamine (6-OHDA)-induced Parkinson's disease(PD) model. Methods Rats were subjected with 6-OHDA in bilateral substantia nigra (SN) as PD model. The location and quantitative detecting techniques inclu-ding immunohisto- chemistry and western blotting were used to determine the expressions and alterations of tight junction proteins (claudin-1, occludin, ZO-1). Results Claudin-1, occludin and ZO-1 were highly expressed in the duodenal mucosa of PD models, where the expressions of occluding (P<0.001) and ZO-1 (P<0.001) were much lower, while the expression of claudin-1 had no alteration. Conclusions The down-regulated expres-sion of occludin and ZO-1 was detected in duodenal mucosa of PD models, which might be related to duodenal ulcer in PD.

ABSTRACT

AIMTo study effects of proanthocyanidins (PA) on contractile activity of isolated aortic smooth muscle in rats and rabbit platelet aggregation.

METHODSIsolated rat aortic muscle rings were adopted to observe the effects of PA on their contraction induced by noradrenaline (NA) or KCl, and their tensions were recorded by BL-310 experimental system of biological function. Rabbit platelet aggregation induced by arachidonic acid (AA), adenosine diphosphate (ADP), and collagen (Coll) was assayed by turbidimetry.

RESULTSPA could significantly inhibit the contraction induced by NA (10(-6) mol/L), low the concentration-response curves of NA and the maximal response on the endothelium-intact or endothelium-denuded aortic rings. But PA couldn't relax the aortic rings precontracted with KCl and had no influence on rabbit platelet aggregation induced by AA, ADP, and Coll.

CONCLUSIONPA can inhibit the contraction induced by NA but not by KCl on isolated rat aortic rings. It also has no influence on rabbit platelet aggregation.

ABSTRACT

OBJECTIVETo analyze the incidence trends of primary liver cancer (PLC) in Qidong.

METHODSData of PLC incidence from 1975 to 1999 in Qidong were analyzed to delineate temporal trends and birth cohort patterns, using age-period-cohort models.

RESULTSSignificant moderation or decreasing trends were began to notice in incidence rates on cohorts born in 1913 - 1917 and 1958 - 1962.

CONCLUSIONResults showed that the incidence risk of the birth cohorts after 1958 - 1962 started to decline. The changes were possibly associated with the implementation of some practical measures on prevention.
